If sending an email-only 499, edit your audience so it only includes people who have an email address on file. This is often best done using an exclusion rule (Email > Does not have a value). This will help the bulk mailing tool process and create mail faster.

Secondly, if performing a series of bulk mailings where you wish to only send to a person once during a series, create a system of issue codes for 499’s and use one issue code per 499 series. Use the issue as an exclusion rule in your main audience, and then be sure to tag that issue whenever creating the bulk mailing. This will tag each of the messages generated in the 499 bulk mailing, and as a result, those 499 people will be automatically excluded from the original audience when you go to create the next 499 bulk mailing.

If you receive an error code or experience long load time for your audience, it may be too complicated and needs fewer rules.
